1592-Skull Bone Marrow Immune Signaling in Chronic Pain
The research article examines how skull bone marrow (SBM) serves as a critical, yet previously overlooked, center for neuroimmune signaling in individuals suffering from chronic pain . By utilizing advanced PET/MRI imaging , researchers discovered that patients with chronic low back pain and knee osteoarthritis exhibit significantly higher levels of TSPO , a protein marker indicating increased immune cell density within the skull. These elevated signals were spatially linked to specific clinical symptoms, including pain intensity, anxiety, and depression , suggesting that the SBM actively communicates with the brain during persistent illness. The study's findings are further reinforced by postmortem tissue analysis , which showed greater cellularity in the marrow of a donor with a history of chronic pain. Ultimately, this work identifies the skull's hematopoietic niche as a potential biological driver of long-term suffering and a novel target for diagnostic and therapeutic intervention .
